Tadpoles of the frog, Xenopus laevis, were exposed to three waterborne concentrations of mefenpyr-diethyl (and control) under flow-through conditions in an extended Amphibian Metamorphosis Assay (EAMA) study design (OCSPP 890.1100, OECD231). Results of the EAMA indicated no specific concerns for thyroid endocrine activity. However, at the highest mean measured test concentration of 0.86 mg a.i./L, tadpoles showed decreased growth metrics a i.e. decreases in both whole-body wet weight and snout-vent length. No adverse effects were noted at the 0.27 mg a.i./L treatment level.

The EAMA is not well-suited to determine No Observable Adverse Effect Concentrations (NOAECs) and Low Observable Adverse Effects Concentrations (LOAECs) based on limited treatment concentrations (n=3) in the study design. It should be noted that in a 28-day rainbow trout growth toxicity study with mefenpyr-diethyl resulted in a NOAEC of 0.1 mg a.i./L based on the observations of slow reaction, food consumption, and staggering movements at concentrations aY= 0.32 mg a.i./L. Thus, the chronic fish NOAEC value is also protective of aquatic phase amphibians.
